From September 2019 to October 2020, pathogenetic analysis of three patients clinically diagnosed as transfusion-related acute lung injury (TRALI) caused by human leukocyte antibodies was conducted by Guangzhou Blood Centre, including 2 males and 1 female, aged 56, 50 and 20 years old, respectively. Solid phase agglutination, anti-human globulin test and flow cytometry method were used to detect the presence of antibodies against patients. Sequencing-based human leukocyte antigen (HLA-SBT) typing technique was used to detect the human leukocyte antigen (HLA) genotypes of patients. Lifecodes single antigen class Ⅰ/Ⅱ kit (LSA-Ⅰ/Ⅱ) were used to detect the specificity of HLA-class Ⅰ and class Ⅱ antibodies in donor blood by Luminex 200 liquid suspension chip system. The HLA specific antibodies and corresponding epitopes in donors were also analyzed. The results showed that HLA class Ⅰ or class Ⅱ specific antibodies against TRALI patients were detected in the blood donors. The plasma of donor 3 received by patient 1 contained antibodies against the patient's HLA-DRB1*09∶01 antigen, and the epitopes mediating the antibody reaction of the donor and recipient were 70R, 31I, 70QA. There were antibodies against the HLA-A*11∶02, HLA-A*11∶01, DRB1*12∶02, and DRB1*09∶01 antigens of patient 2 in the plasma of donor 4, and the associated antigenic epitopes were 151AHA, 57V, and 16Y. Antibodies against the HLA-DRB1*14∶04, DRB1*11∶01, and DPB1*05∶01 antigens of patient 3 were present in the plasma of donor 6 and donor 7, and the associated epitopes were 96HK, 140TV, 13SE, and 111K. Three cases of TRALI were confirmed to be caused by HLA antibodies through laboratory analysis, and human leukocyte antibody detection should be paid attention in clinically suspected cases of TRALI, and targeted diagnosis and treatment should be given.

The (30012) ← (00001) band of 12C16O2 in the 1.6 µm region is used for satellite observation of carbon dioxide in the Earth's atmosphere. Here, we report a Doppler-free spectroscopy study of this band with comb-locked wavelength-modulated cavity-enhanced absorption spectroscopy. Frequencies of 18 transitions with the rotational quantum numbers up to 42 were determined with sub-kHz accuracy, corresponding to a fractional uncertainty at the 10-12 level. With this precision, we revealed an anomalous decrease of the line shift and an increase of the line broadening for the Lamb dips of CO2 in the low-pressure regime compared to values obtained from Doppler-limited spectra at higher pressures.

OBJECTIVE: To understand the distribution of nerve fibers and the types of neural cells in Aspidogaster conchiola. METHODS: Whole worms were subjected to silver staining, histochemical staining and hematoxylin-eosin (HE) staining, and the nervous systems of the worms were observed. RESULTS: There were 3 types of neural cells in the worm head near the cerebral ganglion, including unipolar, bipolar and multipolar neurons, which were divided into 7 types according to the morphology. There was a nerve network on the surface of pharynx and intestinal tract, as well as the reproductive organ, including testis, ovary, lower uterus and penis sac. The nerve network was consisted of circular and longitudinal nerve fibers, and the structure of the nerve network around the mouth was similar to central nerve. CONCLUSIONS: The structure of the A. conchiola central nervous system is very complicated, and the neural networks may be associated with the physiologic activity of the worm. Different neural cells may have diverse functions.

Microsatellites are playing an important role in paternity assignment of animals. Given the cost and effort, it would be optimal to develop a minimal microsatellite marker set for paternity testing. This study was the first to assess paternity in a captive colony of rhesus macaques (Macaca mulatta) from the Chinese province of Anhui using 10 polymorphic microsatellites. Results indicated that if at least 6 loci were genotyped, the probability of paternity assignment success was nearly 100%. Our results provide a panel of 6 markers that is effective for assessing paternity of subspecies M. m. siamica of Anhui origin.

This work studied the destruction of various M-EDTA complexes and trace organic pollutants in treated reusable wastewater under advanced oxidation using UV irradiation and ozonation. Effect of dosage of hydrogen peroxide and acidity of reaction matrices on oxidation efficiencies were investigated. The rate constant of mineralization presents a decreasing trend as Fe(III)-EDTA > Fe(II)-EDTA > Al(III)-EDTA > Pb(II)-EDTA > Na(I)-EDTA > Zn(II)-EDTA > Cu(II)-EDTA. The mineralization efficiencies using ozone alone are 15%, 40% and 15% for the water samples after reverse osmosis (RO), microfiltration (MF) and superfiltration (SF) processes, respectively. The presence of hydrogen peroxide in photochemical reaction matrixes can effectively enhance the mineralization of organic carbon species. When 150 mg l(-1) of H2O2 was added in the effluents, the mineralization markedly increased to 80%, 92% and 89%, respectively.

Plasmid pBR322 and its derivative containing strong promoter phi 10 of bacteriophage T7 RNA polymerase were used as vectors. A fragment of bacteriophage T7 DNA which was digested with two restriction endonucleases (AvaII and HaeIII) was cloned in the BamHI site of plasmid pBR322 and its derivative pAR951, respectively. The inserted DNA is a segment of 632 base pairs containing the complete coding sequence of both T7 gene 3.5 and weak promoter phi 3.8 for bacteriophage T7 RNA polymerase. The function of T7 gene 3.5 is known to code for bacteriophage T7 lysozyme. Transformants that carry the recombinant plasmid were tested for intracellular lysozyme by adding CHCl3. Both cloned strains produce active T7 lysozyme. The gene product, T7 3.5 protein, was analyzed by 10 -20% gradient polyacrylamide- SDS electrophoresis. The result showed that the expression of inserted T7 gene 3.5 in pBR322 derivative is stronger than that in pBR322.

A simple method for restoration of linearly blurred images is described utilizing a weak reference beam, coherence spoiling in one dimension, and a provision for scale change. Experimental results are given.
